Genomics of vasculitis: lessons from mouse models.

چکیده
A genome analysis of mouse models may shed some light on the complex clinicopathological manifestations of systemic vasculitis. In the study of susceptibility loci to vasculitis in MRL mouse models, we found that systemic vasculitis developed through the cumulative effect of multiple gene loci, each of which by itself did not have a significant effect in inducing the related phenotype, thus indicating a polygenic system. The mice developed vasculitis in an additive manner with a hierarchical effect. Some of the susceptibility loci seemed to be common to those in other collagen diseases. Moreover, the loci controlling tissue specificity of vasculitis were present. One of the positional candidate genes for vasculitis showed an allelic polymorphism in the coding region, thus possibly causing a qualitative difference in its function. As a result, a particular combination of polygenes with such an allelic polymorphism may thus play a critical role in leading the cascade reaction to develop vasculitis, and also a regular variation of systemic vasculitis. This is designated as the polygene network in systemic vasculitis. (J Jpn Coll Angiol, 2009, 49: 11-16).

O-2: A Novel Antioxidant Formulation to Treat Male Infertility Emanating from Sperm Oxidative DNA Damage: Promising Preclinical Evidence from Mouse Models
Background: Sperm DNA damage (SDD) is a significant male infertility factor, yet it is not routinely diagnosed or treated in couples undertaking fertility treatment by ART. Men with this condition are likely to experience sub-fertility or infertility, expose their female partners to greater risk of miscarriage and pass on de novo sporadic DNA mutations potentially compromising the health of the...
متن کاملMalignant Glioma: Lessons from Genomics, Mouse Models, and Stem Cells
Eighty percent of malignant tumors that develop in the central nervous system are malignant gliomas, which are essentially incurable. Here, we discuss how recent sequencing studies are identifying unexpected drivers of gliomagenesis, including mutations in isocitrate dehydrogenase 1 and the NF-κB pathway, and how genome-wide analyses are reshaping the classification schemes for tumors and enhan...
متن کاملI-54: New Models for Human and Mouse Genetic
The possibility to reprogram somatic human cells will greatly and deeply change genetic approach and allow the development of new tools to study genetics diseases. Indeed, our ability to study human genetic diseases suffers from the lack of valid in vitro models. The latter should (i) be originating from human primary cells, (ii) be able to self-renew for a long time and (iii) be able to differ...
متن کاملIs there a role for TNF-alpha in anti-neutrophil cytoplasmic antibody-associated vasculitis? Lessons from other chronic inflammatory diseases.
Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis is the most common cause of rapidly progressive glomerulonephritis and immune-mediated pulmonary renal syndrome. Now that the acute manifestations of the disease generally can be controlled with immunosuppressive drugs, ANCA-associated vasculitis has become a chronic and relapsing inflammatory disorder. The need to develop safer ...
متن کاملIn vivo approaches to investigate ANCA-associated vasculitis: lessons and limitations
Anti-neutrophil cytoplasm autoantibody (ANCA)-associated diseases are autoimmune conditions characterized by necrotizing inflammation of small blood vessels. The immunogenesis and etiology of these conditions are unknown, but our knowledge of the immunopathogenesis has increased considerably in recent years. In this review, we discuss the animal models currently used to investigate the mechanis...
